To do this, they measured several inflammatory markers in the blood: substances that can provide information about immune system activity. Around 80 minutes after ingestion, TNF-\u03b1 levels were lower in participants who had received psilocybin. Seven days later, two other markers, IL-6 and CRP, were lower compared with the placebo group.<\/strong><\/p>\n<p><strong>This does not automatically mean that psilocybin is \u201canti-inflammatory\u201d or that it improves the immune system. It does show that a single dose was associated with measurable changes in several substances involved in inflammatory processes.<\/strong><\/p>\n<p><strong>The study was placebo-controlled and published in 2023 in Brain, Behavior, and Immunity.<\/strong><\/p>\n<p><strong>Reference: Mason, N.L., Szabo, A., Kuypers, K.P.C., et al. Brain, Behavior, and Immunity, 114, 299\u2013310.<\/strong><\/p>\n<h2><\/h2>\n<h2><strong>Why researchers are looking into this<\/strong><\/h2>\n<p>Inflammation often sounds like something negative, but that is not necessarily the case. An inflammatory response is a normal and essential part of the immune system. For example, when you have an infection or sustain an injury, inflammation helps your body respond and recover.<\/p>\n<p>It becomes more relevant when inflammatory processes remain active for a prolonged period. Research has shown that, on average, higher levels of certain inflammatory markers are found in some psychiatric conditions, including depression. This is why markers such as CRP, IL-6 and TNF-\u03b1 are increasingly being studied in relation to mental health.<\/p>\n<p>At the same time, researchers are investigating whether psilocybin may have potential effects on conditions such as depression. The Maastricht University researchers therefore wanted to explore whether changes in the immune system could be one of the biological processes that occur after psilocybin administration.<\/p>\n<p>Importantly, this study was not investigating psilocybin as a treatment for inflammation. And because the participants were healthy, the findings cannot be directly applied to people with depression or chronic inflammatory conditions.<\/p>\n<p>&nbsp;<\/p>\n<h2><strong>How was the study set up?<\/strong><\/h2>\n<p>The study included 60 healthy participants. One group received psilocybin, while the other received a placebo. The psilocybin group was given a single dose of:<\/p>\n<ul>\n<li>0.17 milligrams of psilocybin per kilogram of body weight.<\/li>\n<li>For someone weighing 75 kilograms, that equals approximately 12.75 milligrams of psilocybin.<\/li>\n<\/ul>\n<p>This is a psychoactive, psychedelic dose and clearly not a microdose.<\/p>\n<p>The researchers collected blood samples at several points:<\/p>\n<ul>\n<li>before ingestion;<\/li>\n<li>around 80 minutes after ingestion, during the peak of the effects;<\/li>\n<li>seven days later.<\/li>\n<\/ul>\n<p>They measured five substances in the blood that are involved in inflammatory processes:<\/p>\n<ol>\n<li>TNF-\u03b1<br \/>\nA signalling molecule of the immune system that plays an important role in initiating and regulating inflammatory responses.<\/li>\n<li>IL-1\u03b2<br \/>\nA type of cytokine: a small signalling molecule that immune cells use to communicate with each other.<\/li>\n<li>IL-6<br \/>\nAnother cytokine. IL-6 can increase during infections and inflammation and plays a role in several immune processes.<\/li>\n<li>IL-8<br \/>\nA signalling molecule that can help attract certain immune cells to sites of inflammation.<\/li>\n<li>CRP<br \/>\nC-reactive protein. This protein is produced by the liver and is commonly used as a general marker of inflammatory activity in the body.<\/li>\n<\/ol>\n<p>The researchers also assessed mood and social functioning. In a subset of participants, they additionally examined how the body responded to a psychosocial stress test seven days later.<\/p>\n<p>&nbsp;<\/p>\n<h2><strong>What did the researchers find?<\/strong><\/h2>\n<h3>TNF-\u03b1 decreased during the psychedelic experience<\/h3>\n<p>Around 80 minutes after taking psilocybin, TNF-\u03b1 levels were lower than before ingestion and compared with the placebo group. The other inflammatory markers measured did not show a similarly clear decrease at that time. In other words, the effect was not seen across all measured markers at once.<\/p>\n<h3>IL-6 and CRP were lower seven days later<\/h3>\n<p>At the seven-day follow-up, TNF-\u03b1 had returned closer to its original level. At that point, however, the researchers found lower levels of IL-6 and CRP in participants who had received psilocybin.<\/p>\n<p>This is interesting because it suggests that changes in some immune markers may persist beyond the psychedelic experience itself. However, a single measurement after seven days cannot tell us exactly how long these changes last or what their practical significance may be.<\/p>\n<p>&nbsp;<\/p>\n<h2><strong>What does a lower inflammatory marker actually mean?<\/strong><\/h2>\n<p>Lower is not automatically better, because inflammatory responses are essential for healthy functioning. TNF-\u03b1 and IL-6, for example, are not simply \u201cbad\u201d substances. They play several important roles in the immune system. Persistently elevated inflammatory activity, however, can be harmful and is being studied in relation to a range of physical and mental health conditions. It is therefore important to distinguish between:<\/p>\n<ul>\n<li>a temporary change in a single blood marker; and<\/li>\n<li>a lasting reduction in harmful, chronic inflammation.<\/li>\n<\/ul>\n<p>This study shows the first, not the second. Based on these findings, we therefore cannot conclude that participants became healthier simply because certain inflammatory markers decreased.<\/p>\n<p>&nbsp;<\/p>\n<h2><strong>There was also a link with mood<\/strong><\/h2>\n<p>The researchers observed something notable: participants whose IL-6 and CRP levels had decreased more strongly after seven days also reported, on average, more positive changes in mood and social functioning. This is interesting and warrants further research, but it does not prove a direct cause-and-effect relationship. For example:<\/p>\n<ul>\n<li>a change in the immune system may have influenced mood;<\/li>\n<li>another effect of psilocybin may have influenced both mood and immune markers;<\/li>\n<li>or the association may have arisen in another way.<\/li>\n<\/ul>\n<p>Based on this study, we therefore cannot say:<\/p>\n<blockquote><p>\u201cLess inflammation led to a better mood.\u201d We can only say that the two changes were associated with each other.<\/p><\/blockquote>\n<h2><\/h2>\n<h2><strong>The brain was also examined<\/strong><\/h2>\n<p>During the study, researchers used MR spectroscopy to measure glutamate in the hippocampus. Glutamate is one of the brain\u2019s most important signalling molecules. The hippocampus is a brain region involved in functions such as memory, emotion and the processing of experiences.<\/p>\n<p>The acute decrease in TNF-\u03b1 was associated with changes in glutamate levels in the hippocampus. Again, this was an association, so the researchers cannot conclude that one directly caused the other. What makes this particularly interesting is that the study examined changes in several areas at the same time:<\/p>\n<ul>\n<li>the immune system;<\/li>\n<li>the brain;<\/li>\n<li>and the psychological experience.<\/li>\n<\/ul>\n<p>This helps researchers build a better understanding of the possible biological mechanisms underlying the effects of psilocybin.<\/p>\n<p>&nbsp;<\/p>\n<h2><strong>The stress response did not clearly change<\/strong><\/h2>\n<p>Seven days after the psilocybin experience, a subset of participants underwent a psychosocial stress test. The researchers found no significant difference between the psilocybin group and the placebo group in their response to this stressor. This is relevant because an earlier version of the study \u2014 before it had undergone final scientific review and publication \u2014 reported stronger conclusions about a reduced cortisol response.<\/p>\n<p>That effect was not confirmed in the final published study. When reading summaries of scientific research online, it is therefore always important to check whether they refer to a preliminary version or to the final, peer-reviewed publication.<\/p>\n<p>&nbsp;<\/p>\n<h2><strong>What can\u2019t you conclude from this study?<\/strong><\/h2>\n<p>This is perhaps the most important part of the study. This was not a study on microdosing, but on psilocybin at a much higher dose. Participants received approximately 0.17 milligrams of psilocybin per kilogram of body weight. This is a psychedelic dose that produces clear subjective effects.<\/p>\n<p>A microdose is considerably lower and is specifically intended to avoid a full psychedelic experience. You therefore cannot assume that a microdose produces the same effects, simply on a smaller scale. Biological effects do not always increase or decrease proportionally with dose. This study therefore does not show that microdosing lowers IL-6, CRP or TNF-\u03b1.<\/p>\n<p>&nbsp;<\/p>\n<h2><strong>Lower levels do not automatically mean a healthier immune system<\/strong><\/h2>\n<p>The immune system is not a volume dial that simply needs to be turned down. In some situations, you actually need a strong inflammatory response, for example when fighting an infection. A change in a few inflammatory markers therefore does not automatically mean that someone has developed a stronger, better or healthier immune system. To establish that, research would need to look at meaningful health outcomes, rather than changes in individual blood markers alone.<\/p>\n<p>&nbsp;<\/p>\n<h2><strong>The participants were healthy<\/strong><\/h2>\n<p>The participants did not have depression, an autoimmune disease, or another condition involving elevated inflammatory activity that required treatment.<\/p>\n<p>The study therefore does not tell us what psilocybin does in people who already have increased levels of inflammatory markers. It also does not show whether the changes observed in these markers are clinically meaningful.<\/p>\n<p>&nbsp;<\/p>\n<h2><strong>An association does not prove cause and effect<\/strong><\/h2>\n<p>The decrease in IL-6 and CRP was associated with more positive changes in mood. However, this does not tell us whether the change in inflammatory markers actually caused the psychological change. Establishing a cause-and-effect relationship would require further research.<\/p>\n<p>&nbsp;<\/p>\n<h2><strong>One study is not enough to draw a conclusion<\/strong><\/h2>\n<p>Scientific findings become more reliable when different research groups are able to reproduce the same effect. For these specific findings, there is still limited replication research available. Other studies on psilocybin and the immune system have also not found exactly the same pattern of results.<\/p>\n<p>&nbsp;<\/p>\n<h2><strong>Other studies have found different results<\/strong><\/h2>\n<p>The study by Mason and colleagues does not stand alone. Other research shows why it is still too early to conclude that psilocybin has an anti-inflammatory effect. For example, a smaller human study found no significant changes in several inflammatory markers one day after a single dose of psilocybin.<\/p>\n<p>Another analysis combined data from 91 participants across three clinical trials. In the pooled data, IL-8 was temporarily elevated within one week after psilocybin. However, when the individual studies were analysed separately, these changes were not statistically significant.<\/p>\n<p>Preclinical research, for example in cells and animals, also suggests that the effect may depend on the circumstances. An immune system that is already in an inflammatory state may respond differently from that of a healthy person.<\/p>\n<blockquote><p>So the picture is not yet: psilocybin reduces inflammation. A more accurate conclusion is that psilocybin appears to interact with processes within the immune system, but how, when and in whom is still not sufficiently understood.<\/p><\/blockquote>\n<h2><\/h2>\n<h2><strong>Why do we still find this study interesting?<\/strong><\/h2>\n<p>Psilocybin research often focuses primarily on what people experience: changes in mood, perception, emotions or behaviour. This study shows that researchers are also increasingly trying to understand what is happening biologically in the body at the same time.<\/p>\n<p>The notable finding is that a single psychedelic dose was not only associated with a change in TNF-\u03b1 during the experience, but that differences in IL-6 and CRP were also found seven days later. That makes it an interesting lead for further research. But it is not yet evidence that psilocybin treats inflammatory diseases, nor is it evidence that microdosing improves the immune system.<\/p>\n<p>&nbsp;<\/p>\n<h2><strong>What does this mean for microdosing?<\/strong><\/h2>\n<p>For now, it mainly leaves an important question unanswered. If a psychedelic dose of psilocybin can influence certain immune markers, does something similar happen with much lower, repeated doses? That is scientifically interesting, but this study does not provide an answer. To investigate this properly, researchers would need to follow participants throughout a microdosing protocol and measure inflammatory markers before, during and after the protocol, while comparing the results with a placebo group. Until that kind of research is available, the findings of this study cannot be directly applied to microdosing.<\/p>\n<p>&nbsp;<\/p>\n<h2><strong>The bigger picture<\/strong><\/h2>\n<p>Research into psychedelics and the immune system is still at a relatively early stage. There are indications that psilocybin and other psychedelics may influence immune processes, but studies differ in dose, study design, participants and outcomes.<\/p>\n<p>Some studies find decreases in certain markers. Others find no change. And still others find temporary increases. That is not unusual in a young field of research.
